Week in Montalcino

My husband and I went to a charity auction and bid on -- and won! -- a week in a Montalcino villa. We love Italy but we`ve been strictly train travelers until now. This will push us to finally rent a car (in Siena, I`m thinking) and start exploring the areas that aren`t accessible by train. We are doing our research on the wineries, but we won`t want to limit ourselves to just wineries and Montalcino. What are some reasonable day trips we could take?
I`d like to spend a little time in Florence again, and a day trip would be perfect if you don`t think the timing is too exhausting. What else? Our favorite Tuscan/Umbrian towns before were Lucca and Perugia, to give you some perspective.
